Technical Abstract: A new LC/MS-MS method for analysis of allelopathic chemicals in Rye was developed. Methoxy-substituted forms of the hydroxamic group were re-discovered along with measuring the main hydroxamic moieties, DIBOA and BOA, and small amounts of their precursors, the glucosylated forms. Samples were also examined for Beta-phenylactic acid (PLA) and Beta-hydroxybutyric acid (HBA), and small amounts were measured. The preparation method involved aqueous incubation of the dried/ground plant materials (1:15 wt/wt, plant:water) for 12 hours which dictated that a greater quantity of the aglucones would be present since the plants have natural levels of Beta-glucosidases that degraded the glucosylated forms. The analytical method is reproducible, is sensitive, and requires no sample cleanup and very simple preparation procedures. MDLs were as follows: hydroxybutyric - 15 ng/g, phenylacetic - 3.4 ng/g, DIBOA-glucose - 2.8 ng/g, DIBOA - 1.6 ng/g, BOA - 3.0 ng/g, DIMBOA-glucose - 10.5 ng/g, DIMBOA - 0.3 ng/g, MBOA - 2.13 ng/g.
