Interpretive Summary:

Technical Abstract: The genetic variability among 17 isolates of Sclerotium rolfsii from Oklahoma and other locations was investigated using mycelial compatibility groups (MCGs) and Random Amplification Polymorphism DNA PCR (RAPD-PCR). Pairings among isolates of S. rolfsii on potato dextrose agar containing 100 micro g/ml of streptomycin sulfate resulted in 14 different MCGs. Isolates from Oklahoma were placed in three different MCGs, and the remaining isolates were placed in individual MCGs. RAPD-PCR analyses revealed similarities in banding patterns of isolates belonging to the same MCG with primer 335. Four distinct groups of isolates were identified based on DNA polymorphisms identified by RAPD-PCR using 21 primers. Isolates were clustered together according to their host origin rather than by their geographic location or year of recovery. These results indicate that MCG and RAPD-PCR analysis are valuable methods for differentiating the genetic variability of S. rolfsii isolates.
