Interpretive Summary:

Technical Abstract: A standardized laboratory scale procedure to measure ethanol yield from corn has been developed in a multi-institutional project. The process was developed to provide a reference procedure, available in the public domain, for measurement of ethanol produced from corn. The procedure uses 25 g of corn, ground through a hammermill prior to liquefaction using commercially available alpha amylase. Commercially available glucoamylase and yeast are added for simultaneous saccharification and fermentation (72 hr). HPLC and gravimetric techniques were used to measure ethanol yield. The procedure was evaluated for variability in ethanol yield by repeated testing of a single hybrid; the coefficient of variability for ethanol yield was less than 5%. Ethanol yields of a broad range of diverse hybrids were tested to verify the sensitivity of the procedure and will be presented. The usefulness of the procedure as an industry benchmark and future research using the procedure will be discussed.
