Publication Type: Abstract Only Publication Acceptance Date: 7/3/2001 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: The A. thaliana J-domain molecular chaperones comprise an unexpectedly large and complex family of proteins. In silico analysis of the localization of all 89 J-domain proteins resulted in predictions that various species are present in each of the subcellular compartments. Surprisingly, it was predicted that 27 of the 89 J-domain proteins are located in the nucleus. We are now conducting experimental analysis of the localization predictions. No counterpart of the atDjC6 protein has yet been identified in yeast or mammalian systems. The atDjC6 sequence contains three potential Nuclear Localization Signals (NLS). The actual subcellular localization of atDjC6-GFP, transiently expressed in tobacco cells is being determined by confocal microscopy. The NLS sequences will be evaluated, both individually and in combination, by site-directed mutagenesis, in order to determine which is necessary and sufficient for nuclear localization. Both the wild-type and mutant atDjC6 proteins will be tested for interaction with importin-a. |
