Interpretive Summary:

Technical Abstract: Dioecious Ilex present a unique plant model to search for sex related molecular markers. Female I. Crenata Sky Pencil cv. and male I. Crenata microphylla and their F1 progeny were analyzed for the presence of AFLP markers associated with sex determination. Although little is known about the genetic and physiological basis for sex determination in this genus, the use of modern molecular marker techniques may provide tools for studying the mechanism of sex determination, as well as assist in breeding programs. We present here male and female specific AFLP markers. There were six PCR amplified unique DNA bands. They were purified, cloned onto pT-Adv Vector, sequences and specific genomic primers were designed for PCR-based sex identification in seedlings at a vegetative stage.
