Technical Abstract: Genes which encode for cytokines are potential candidates for disease resistance QTL in cattle, however relatively few members of this gene family have been positioned on the bovine genetic map. To improve upon this condition, we determined the cytogenetic map locations for seven cytokine genes that are specifically involved with either growth and differentiation of T-lymphocytes or the immune response initiated by T-cells to generate effector cell populations. Unique bovine cDNAs were cloned for IL10, IL12-p35, IL12-p40, IL15, IL18, TNF-alpha, and TGF-beta; and radiolabelled probes made from these clones were used to screen a bovine BAC library. FISH probes generated from these BAC clones were used to determine physical assignments. Localized FITC signals were detected at BTA16q1.2 for IL10, BTA1q3.4-3.6 for IL12-p35, BTA7q1.4-1.5 for IL12-p40, BTA17q1.3-1.4 for IL15, BTA15q1.3-1.4 for IL18, BTA23q2.1-2.2 for TNF-alpha, and BTA18q2.4-2.5 for TGF-beta. Alignment of bovine and human maps based on these data agreed with previous ZOO-FISH results with respect to comparison of conserved synteny. Currently, microsatellite markers are being developed from these assigned, non-chimeric BAC clones to position these genes on the bovine linkage map. Informative markers associated with these cytokine gene will be incorporated into a genome wide QTL scan of a resource cattle population with defined levels of heritable resistance to parasitic nematode infection.
