Technical Abstract: This study characterizes the temperature sensitivity of AtTS02, an Arabidopsis thaliana acquired thermotolerance mutant. Exposed to etiolated Arabidopsis seedlings to 48 or 50 deg C for 30 min blocks Chl accumulation upon subsequent exposure to light at 25 deg C. However, a milder treatment, 44 deg C for 30 min, simply delays Chl accumulation and is not lethal. Arabidopsis seedlings can be protected against the effects of the 44 deg C 30 min challenge by a 4 h preincubation at 38 deg C. By the use of the milder challenge and protective pretreatment, mutants lacking components of the acquired thermotolerance system can be isolated. The phenotype of such mutants would be a delayed accumulation of Chl in the light at 25 deg C after exposure to the sublethal 44 deg C treatment. Analysis of seedlings derived from selected mutants revealed reduced levels of protection compared with control seedlings. Characterization of the acquired thermotolerance mutant AtTS02 showed reduced high temperature stress protection. Genetic analysis showed that the loss of acquired thermotolerance in AtTS02 was a recessive trait.
