Technical Abstract: Wheat germplasm is susceptible to barley yellow dwarf virus (BYDV). A wheat alien substitution line was developed that contains a Thinopyrum intermedium 7E chromosome conferred resistance to BYDV. To minimize the amount of alien chromatin in this alien substitution line, translocation lines were developed by subjecting monosomic alien substitution line seeds to gamma-irradiation. Six resistant translocation lines, which were shown by molecular marker analysis to contain less than a complete Thinopyrum intermedium 7E chromosome, were selected for developing elite wheat cultivars resistant to BYDV. These translocation lines were advanced to the BC2 generation with wheat as the recurrent parent. A set of 534 BC2F1 plants were inoculated with BYDV and following a two-week incubation period assayed by ELISA for BYDV resistance. Thirty-eight percent of these plants were resistant to BYDV. Resistant and susceptible subsets of these BC2F1 plants were analyzed molecularly using markers previously positioned on either the short and long arm of the 7E chromosome. The results of the marker analysis will show the segregation and estimate the size of the Thinopyrum intermedium chromosome segment(s) contained in the BC2F1 wheat lines.
