Technical Abstract: A candidate-gene approach was applied to isolate resistance gene analogs from a potato hybrid line of Solanum phureja x S. stenotmum with late blight resistance using degenerate primers based on conserved motifs within the nucleotide binding site of cloned resistance genes. PCR-amplified products of approximately the predicted size (500 bp) were purified from the agarose gel and cloned with a PCR cloning kit. Individual clones were grouped by restriction analysis and one representative from each group was sequenced. DNA sequence comparisons by the BLAST search showed that some of the isolated clones were highly similar to resistance genes from other plant species and they were highly similar to resistance genes from other plant species and they were thus classified as resistance gene analogs (RGA). These RGAs, when used as probes in Southern hybridization, detected polymorphism between the late blight resistant and susceptible parental lines, which were used to construct an F1 mapping population. Genetic analysis of these RGAs in relation to late blight resistance in potato is in progress.
