Technical Abstract: Lipoxygenase in potato tubers is believed to play an important role in defense mechanisms. Thirteen potato cultivars reported to have late blight resistance were examined for severity of blight on foliage and tubers, total protein, lipoxygenase specific activity and 5-LOX protein. Crude protein extracts were prepared with potassium phosphate buffer and subjected to centrifugation. The lipoxygenase specific activity was measured by polarographic method using arachidonic acid as an enzyme substrate. 5-LOX protein was estimated through ELISA and western blot. Foliage reaction was assessed under field conditions but tubers were inoculated in the lab. There were significant differences among cultivars for both foliage and tuber disease severity, but there was no correlation of foliage with tuber disease severity. Cultivars differed significantly for total protein, lipoxygenase specific activity and 5-LOX protein. These were not correlated with late blight reaction under our evaluation methods.
