Interpretive Summary:

Technical Abstract: VPI of germinating kernels with MDMV resulted in mosaic symptoms on the first 4-6 leaves of resistant Pa405 (R) and Oh28 susceptible (S) inbreds, whereas later appearing leaves of S but not of R maize showed mosaic. To determine the stage when MDMV infection is arrested in R plants, the progression of infection was followed in germinating kernels after VPI. Kernels were inoculated with MDMV 6 hr after the start of germination and incubated for up to 3 days at 30O C. Tissue prints of excised embryos probed with MDMV antiserum showed little or no MDMV in S and R embryos at 0 and 18 h after VPI, whereas by 42 h MDMV occurred in discreet areas of the scutellum and embryo axis. After 64 h, high MDMV titers were present in large but still discreet areas of the embryo. The distribution of MDMV indicated the presence of virus in or near the vascular bundles of the scutellum and embryo axis. No virus was detected in mock-inoculated embryos. The similar virus distribution in R and S embryos indicated that MDMV infection was not arrested at this early stage in R maize.
