Technical Abstract: Strain A-145 is a mini-Tn5 Km mutant of the potential bacterial biocontrol agent Enterobacter cloacae strain 501R3. Populations of strain A-145, applied as a seed treatment, did not increase in numbers over a 45 h incubation period in cucumber or wheat spermospheres, whereas strain 501R3 increased 13-fold and 600-fold, respectively. Colonization by strain A-145 was significantly (P < 0.05) reduced relative to strain 501R3 in cucumber and wheat rhizospheres. Suppression of damping-off of cucumber caused by Pythium ultimum was also significantly (P < 0.05) reduced with strain A-145, relative to strain 501R3. The mutation in strain A-145 resulted in the inability to grow on M56 minimal medium. Colonization and biocontrol both appear to be affected by a single transposon insertion in strain A-145 in contrast to other colonization mutants of E. cloacae.
