Interpretive Summary:

Technical Abstract: Quantitative trait loci (QTL) affecting conformation traits were identified using analysis of variance (ANOVA) to detect associations between the predicted transmitting abilities and marker genotypes of seven large US Holstein grandsire families. Results from a single grandsire family composed of 79 sons indicated strong evidence (P = 0.000021) for an association between dairy form and a microsatellite marker located near the telomere of bovine chromosome 27 (BTA27). Phenotypic scores for dairy form are based upon femininity, conditioning and dairyness, and are moderately correlated with milk production. In order to verify our initial result and proceed with further genetic analysis, 12 additional markers previously placed on linkage maps of BTA27 and OOV26 were tested for heterozygosity in this sire family. Four of these markers were informative. ANOVA analysis of these genotypes supported an association between dairy form and telomeric markers of BTA27. A more exact genetic location of this QTL is being determined by marker interval analysis that includes genotypes generated from single nucleotide polymorphisms of genes identified to lie near the BTA27 telomere by comparative mapping.
