Technical Abstract: Monoclonal antibodies were produced to epizootic hemorrhagic disease virus serotype 2 (EHDV-2) and used to characterize antigenic determinants on the highly conserved VP7. Immune precipitation and binding assays indicated two epitopes; one unique to VP7 of EHDV-2 and one shared between EHDV-1 and EHDV-3. These epitopes are unique to EHDV and are not shared with the U.S. serotypes of BTV. The physical location of the epitopes was examined by comparison of the VP7 sequences from EHDV-1 and EHDV- 2 and computer prediction of potential antigenic sites on VP7. Fine mapping of the epitopes was performed by cloning and expression of truncated VP7 peptides and epitope scanning with synthetic peptides, using Western blotting, immune precipitation, and competitive ELISA. The potential diagnostic application of the monoclonal antibodies defining these epitopes was ascertained in both competitive and antigen capture ELISA.
