Interpretive Summary:

Technical Abstract: In the past several years, subgroup J avian leukosis viruses (ALV-J) have been isolated from commercial broiler breeder flocks experiencing myeloid leukosis in the United States, and are a significant economic concern to the poultry industry. We have been interested in analyzing ALV-J strains to determine the degree of sequence variation between different field isolates, to identify region(s) of the viral genome important in specifyin myeloid tumor induction, and to map the common integration site(s) in ALV-J induced tumors. To this end, we have obtained two field isolates of ALV-J (strains ADOL-Hcl and ADOL-R5-4) and have begun molecular characterization of their genomes. Restriction enzyme mapping and partial sequence analysis have demonstrated that both the ADOL-Hc-1 and ADOL-R5-4 strains contain the E element highly related to that of HPRS-103 the prototype of ALV-J. Portions of the LTR sequenced from the US ALV-J isolates are also similar but not identical to that of the HPRS-103 strain. Interestingly, while on isolate appears to have an env gene highly related to HPRS-103, the other contains restriction enzyme sites more typical of those seen in other common ALV/RSV isolates. This result suggests that different strains of ALV-J have arisen independently and/or are continuing to undergo recombination and selection in the field. These data, as well as functional studies on the LTR and E region, will be presented.
