Interpretive Summary:

Technical Abstract: Resources: An EcoRI restriction fragment length polymorphism (RFLP) of nebulin (NEB) with three variant bands has been reported in crosses of Meishan and Yorkshire swine. A total of 94 individuals from eight two- generation backcross families in the USDA-MARC Swine Reference Population was used in this study. The human cDNA NEB probe was obtained from ATCC, MIM #161650. Procedures used in transformation, amplification and isolatio of cDNA probes were as described. The cDNA was labeled with [32P]-#-ATP to specific activity of 1-2 x 10 counts per minute per microgram (cpm/#g) by random-priming. Results: The RFLP identified among USDA-MARC reference population pigs was characterized by polymorphic fragments of 5.7, 5.8, and 6.1 kilobases (kb) and one constant fragment, 4.7 kb. Although the band patterns were similar, fragment sizes were smaller than reported in Meishan and Yorkshire crosses. All offspring of five reference population families were genotyped to yield 106 informative meiosis. Two-point and multipoint linkage analyses were used to analyze the segregation data & recombination rate computed against over 1,000 existing loci in the USDA-MARC genome database. NEB was assigned to a linkage group based on LOD > 3. Relative mapping order and distance was further tested within the group using the FLIPS and FIXED options. The CHROMPIC option of CRIMAP was used to identify double crossovers and check for genotyping errors. We were able to map NEB to the centromeric end of chromosome 15 (Fig. 1) with the existing USDA-MARC swine map as a reference. This assignment improves our coverage SSC15 as it extends our linkage group by 12.0 cM. NEB maps to human chromosome 2q24.1-24.2, but is not physically assigned in swine.
