Publication Type: Abstract Only Publication Acceptance Date: 12/1/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: Integration of plasmid DNA carrying two marker genes was investigated in transgenic oat (Avena sativa L.) tissue cultures and plants that had been transformed by microprojectile bombardment. Ninety percent of the transformation events had at least one intact copy of one or both of the transgenes. Only in 3% of the tissue cultures and plants was one of the transgenes entirely missing. The intact transgene copies were accompanied by multiple truncated and rearranged fragments of transgenic DNA resulting in complex transgene integration patterns as indicated by the presence of several different size restriction fragments on Southern blot analysis. Complex transgene integration patterns suggested that transgene delivery to the plant cells and/or genomic integration was associated with extensive rearrangement and fragmentation of introduced DNA. All introduced transgene fragments were integrated into one genetic locus in the plant genome as indicated by transgene segregation patterns in the progeny. A possible mechanism of transgene integration resulting in such an arrangement of transgenic DNA will be proposed. |
