Due to a reduction in quality by 1RS in hard wheats and the potential for the same in soft wheats, it is desirable to limit the amount of rye chromatin present in translocations. Soft red winter wheat cultivars, 'Grant', 'Pioneer 2510', 'Freedom', and 'GR 876', were compared to 'Kavkaz', a hard wheat and the source of 1RS.1BL, for amount of rye chromatin. All varieties exhibited the same w-secalin bands when seed endosperm proteins were extracted and separated on a polyacrylamide gel. Fluorescence in situ hybridization to root-tip chromosomes revealed that a repetitive rye telomere probe (pAW 161) hybridized to translocation chromosomes of each variety. Additionally, a dispersed rye repetitive sequence (R173) hybridized to the entire 1RS arm of each variety. Although 'Grant', 'Pioneer 2510', 'Freedom', and 'GR 876' were developed independently by three breeding programs, they all appear to have the same amount of rye chromatin and a stable, whole-arm rye chromosome translocation introgressed from 'Kavkaz'. |
