Technical Abstract: A shriveled seed mutant T311 was found among progeny of a cross between P2180 and AP2. Shriveled seeds are generally small and have puckered/dimpled seed surface. To demonstrate the association between storage protein and the shriveled seeds, plants of T311 and P2180 were grown at specific temperatures allowing expression of the shriveled seed phenotype. Comparative analyses between T311 and P2180 showed that total protein content was decreased in the shriveled seeds. Appearance of alpha and alpha' subunits of conglycinin was delayed in shriveled seeds of T311. SEM analyses showed that mature shriveled seeds had larger protein bodies than wild type seeds. Protein bodies in shriveled seeds of T311 had more scattered globoid crystals and fewer globoid regions than P2180 seeds. Energy disperse X-ray analyses detected the presence of elements P, Ca, Mg in the globoid crystals of T311 shriveled seeds while in P2180 round seeds, elements K and S were observed in addition to elements P, Ca, Mg.
