Interpretive Summary:

Technical Abstract: We have expanded our earlier study of markers linked to the polled locus (Mamm. Genome 1995 6:710-712), to include Simmental and Limousin cattle, in addition to Charolais. Six more markers have also been added. Fifteen paternal half-sib families with a total of 168 calves segregating for horned/polled were tested with 8 microsatellites. AGLA17 was not polymorphic in any of our sires (marker courtesy of ABS). TGLA49 (LOD=12.15) and BM6438 (LOD=10.76) were both linked to polled at 0.1 cM. BMS1928 was linked at 3 cM (LOD=6.81); BM8139 at 9 cM (LOD=4.41); and BMS574 at 21 cM (LOD=3.11) and BMS711 (LOD=3.02) at 22 cM (markers courtesy of USDA, Clay Center). In addition, a deletion in IFNAR was used as a marker and appears to be linked to polled at 9 cM (LOD=3.15). We further attempted to use comparative mapping data to find other markers. KAP8, a marker from sheep 1q, was linked at 11 cM (LOD=3.47). Since this segment of cattle chromosome 1 is homologous to the terminal end of human chromosome 21 we tested human microsatellites but found either no product (HMG14) nor if we detected a product, found no polymorphism (S171).
