Technical Abstract: Forty seven Phomopsis isolates and nine soybean fungal pathogens were tested for genetic diversity by PCR and RFLP with six enzymes. The DNA region of the internal transcribed spacer (ITS) was amplified by ITS4 and ITS5 primers. The amplified DNA of the nine pathogens had different fragment lengths that ranged from 500 to 800 base pairs, while the Phomopsis isolates had a uniform fragment length of around 500 base pairs. Digestion with restriction enzymes revealed distinct band patterns among the various soybean pathogens and among the Phomopsis isolates. Diaporthe and Phomopsis were distinguished by PCR-RFLP in the ITS region. All soybean Phomopsis isolates from different locations showed the same band patterns.
