Interpretive Summary:

Technical Abstract: Inoculum from a barley (Hordeum vulgare) plant presumably infected only with the High Plains pathogen (HPP) was used for vascular puncture inoculation (VPI) studies. Sweet corn (Zea mays) var. Spirit seeds (600) soaked in water for two hours were inoculated 2-3 times with VPI in the vascular bundle area beneath the embryo. Chlorotic streak and spot symptoms developed, beginning on the first leaf, over a 3-wk period after inoculation. Of the plants (53/590) that developed symptoms; 4 and 10 plants tested positive by bioassay and ELISA, respectively, for WSMV. However, 4 plants that developed symptoms at 21 days after inoculation tested positive for HPP by ELISA; 1 of 4 plants tested positive for WSMV. Necrosis of the lower leaves also occurred in these plants. Inoculum from the three HPP- infected plants was not infectious by rub inoculation to either Oh28 corn, Monon wheat, or Atlas sorghum. Studies are underway to confirm mite transmission and further characterize the etiology of this disease.
