Interpretive Summary:

Technical Abstract: Gizzerosine is a toxic compound formed during the commercial production of fish meals which induces black vomit. A homologous radioimmunoassay (RIA) for the detection and quantification of gizzerosine activity in commercial fish meals has been developed. Polyclonal antibodies were produced in rabbits by immunization with gizzerosine conjugated to keyhole limpet protein. The antibody employed in the RIA has a lower detection limit of 0.78 ng/mg and crossreactivity with histamine of 26%. No crossreactivity with histidine or lysine was observed. A mild buffer extraction procedure was used for extracting gizzerosine from fish meal, resulting an average recovery of efficiency of 98%. Displacement curves from extracted and serially diluted fish meal samples were parallel with gizzerosine standard. The RIA was used to determine gizzerosine activity in a pool of 23 fish meal samples of known gizzerosine scores and histamine content as determined by a chick bioassay. The partial correlation coefficient between gizzerosine content as determined by RIA and gizzerosine scores from the bioassay was 0.83 (P<0.01). There were significant correlations between gizzerosine scores and histamine content of the fish meals (0.63) and between histamine content and gizzerosine levels determined by RIA (0.59). The application of the RIA for the determination of gizzerosine activity in commercial fish meals could be of importance for the prevention of gizzard erosions and for studying gizzerosine- induced pathology and metabolism.
