Technical Abstract: Identification of complimentary DNAs (cDNAs) with the corresponding phenotypic/morphological mutants will provide a resource necessary to begin to unravel the complexities of the maize genome. More than 1000 maize mutants have been identified and characterized to varying degrees. Of these, less than 200 have been localized further than chromosome arm. F2 mapping populations were constructed from crosses of individuals containin the mutant allele(s) to maize inbred lines A619, A632, B73 and/or Mo17. F2 individuals or F2-derived F3 families were phenotypically classified and tissue samples collected. Southern analysis of F2 or bulked F3 families was performed using the UMC Maize Core Marker Probe Set on DNAs digestedwith EcoRI and HindIII. Mutants were located to "bins" of the UMC Maize RFLP Map by recombination order using MAPMAKER. The results indicate that this approach can: 1) identify potential cDNA/mutant associations, 2) )indicate possible allelic pairs of mutants, and 3) provide a rapid and cost-effective means of gene localization.
