Interpretive Summary: This study has been conducted to identify interleukin-2 receptor gamma gene which is important for maintaining immune system and best known for causing "Bubble boy disease" in human when a defective gene is present. We isolated the interleukin-2 receptor gamma gene from cattle and showed how this gene is organized such a way that which part of the gene is carrying messages actively translated into the protein and which is not. There are 29 different chromosomes in cattle along with X and Y chromosomes. We showed that there is only one copy of the gene in entire cattle chromosomes. In addition, we determined that the interleukin-2 receptor gamma gene is located on cattle chromosome X. The present study will help to better understand the role of interleukin-2 receptor gamma gene.

Technical Abstract: Interleukin-2 receptor (IL-2R) gamma chain, a member of the cytokine receptor superfamily, forms a high affinity receptor with IL-2R alpha and beta chains which plays an important role in interlelukin-2 (IL-2) signal transduction. We have cloned and characterized the bovine IL-2R gamma gene and corresponding cDNA. Bovine IL-2R gamma is a single copy gene that contains 8 exons and spans approximately 3.8 kb. The promoter region lacks conventional TATA and CCAAT consensus sites, but contains several regulatory elements which are recognition sites for GATA binding proteins, AP-1 and AP-2. Physical assignment by fluorescence in situ hybridization (FISH) placed the bovine IL-2R gamma gene on chromosome Xq23.
