Technical Abstract: The discovery of individual maize chromosome retention in oat genomes after oat x maize crosses and the ability to produce maize chromosome addition oat lines may lead to isolation of chromosome specific maize DNA. This is a key step for: 1. isolation of chromosome specific probes for genome analysis; 2. development of chromosome specific paint (a complex mixture of fDNA probes which hybridize to the entire length of a specific chromosome); 3. dissection of maize chromosomes and physical mapping of the maize genome; and 4. selective isolation of novel cDNAs encoded by the chromosome specific DNA. The goal of this project is the isolation of maize chromosome specific cosmid clones from cosmid libraries constructed from genomic DNA of maize oat chromosome addition lines. To identify clones carrying maize DNA fragments, we proposed to use species specific dispersed nucleotide sequences. Cosmid libraries from DNA of the parental strains of foat (Starter 1) and maize (Seneca 60) and from the maize oat chromosome addition line carrying the maize chromosome 9 were constructed in the pWE15 cosmid vector after partial digestion with Sau 3A and dephosphorylation. We found that both total genomic maize DNA and maize specific repeated sequences hybridized strongly to the homologous cosmid library and showed poor hybridization to clones of the heterologous library. We hope to isolate clones belonging to a specific maize chromosome.
