Technical Abstract: We have carried out, and review here, studies to probe the structure of the active site of the putative receptor involved in dormancy maintenance in wheat embryos. Our approach is to design and test ABA analogues to probe the ABA response system. We anticipate that, in addition to defining the steric and electronic requirements of the ABA response system in dormancy maintenance, the structure/activity results in embryo dormancy could be related to the requirements of other ABA-inducible systems. As well, we hope to identify sites on the ABA molecule that could be used for attachment of photoaffinity and other labels. We have found that analogues are useful tools for relating maintenance of dormancy to expression of a subset of ABA-inducible genes. |
