One way to determine if a sample contains one of these organisms is to look for characteristic signatures in the bacterial DNA. The Polymerase Chain Reaction (PCR) is a specialized method that is able to detect even small numbers of the target cell. This protocol describes the procedures required to detect E. coli and Salmonella using standard PCR methods and detection via a process called gel electrophoresis. Technical Abstract: Purpose To describe the correct procedures on how to set-up polymerase chain reaction (PCR) and run gel electrophoresis for the detection of Escherichia coli (E. coli) and Salmonella. Scope This protocol describes the procedures used to detect the presence of the invA gene from Salmonella and the uidA gene from E. coli using standard PCR, with detection via gel electrophoresis. |
