Interpretive Summary:

Technical Abstract: The immature fiber (im) mutant is one type of cotton fiber mutant with unique characteristics of non-fluffy cotton bolls. Compared to its near-isogenic line (NIL) wild type Texas Marker-1 (TM-1), im fiber has a thin secondary cell wall and is less mature. In this study, attenuated total reflection Fourier transform infrared (ATR FT-IR) spectroscopy was examined to discriminate underdeveloped from developed NIL fibers grown over four years (2007, 2011, 2014, and 2015). Developed NIL fibers were harvested from naturally open bolls in the cotton plant, whereas underdeveloped fibers were collected from unopened bolls in which fibers were still growing at various developmental stages. Results from principal component analysis (PCA) and three algorithm approaches showed a clear separation between underdeveloped and developed fibers and also a separation of NILs within either underdeveloped or developed fibers. Because ATR FT-IR method enables the measurement of breeder’s fiber samples at a small amount in a rapid and non-destructive way, it offers a potential fiber phenotype screening measurement.
