Interpretive Summary:

Technical Abstract: A total of 1,062 apple EST-derived simple sequence repeat (SSR) markers were screened and evaluated for cross-species transferability onto two different pear cultivars. PCR amplification of pear DNA leaf tissue samples was successful using 721 primer pairs demonstrating a high efficiency of transferability (67.9 percent) between apple and pear. Eight markers were selected to assess available genetic diversity in 48 genotypes of pear (Pyrus). A total of 140 gene loci were identified, of which 129 (92.14 percent) were polymorphic. Moreover, different pear cultivars could be readily distinguished. Based on polymorphisms revealed by the polymorphic SSR marker, an Unweighted Pair Group Method with Arithmetic mean (UPGMA) cluster analysis was performed using soft NTY-SIS-pc2.01. Pear genotypes were classified into two major classes and grouped based on their Asiatic and European origin into clusters.
