Interpretive Summary:

Technical Abstract: The "doubled haploid" project started in October, 2010, covering four main approaches: anther culture, microspore culture, use of foreign pollen as inducer, and induced mutation. A set of genetically diverse cultivated lines, wild Helianthus accessions and interspecific amphiploids have been established to provide source material for both anther and microspore culture. Various protocols are being evaluated for their culturing suitability. Cross combinations between cms line and several cultivated lines are being made and the cms F1 plants will be pollinated by pollen of H. tuberosus and safflower accessions in the following season to observe any embryo formation and haploid induction. For the induced mutation approach, pollen of a selected cultivated line will be irradiated by X ray, and used to pollinated onto a selected nuclear male-sterile line in the spring season. The hybrids will be pollinated by a yellow-leaf line this, and the cross progenies will be grown this summer to detect any haploid resulted from mutation.
