Interpretive Summary:

Technical Abstract: It has been previously reported that avian retroviruses, i.e. avian leukosis virus (ALV) and reticoloendotheliosis virus (REV), integrate in the Marek’s disease virus genome affecting MDV pathogenicity. RM-2 is an attenuated serotype 1 MDV virus generated by insertion of the REV LTR in the genome of JM, a virulent MDV strain. Using recombinant DNA technology and classical homologous recombination we generated a recombinant CVI988/Rispens (serotype 1 vaccine strain) virus containing the LTR from RM-2 (CVRM2). This virus replicated to higher levels than CVI988 both in vitro and in vivo and conferred protection equal or superior to CVI988 when challenged with a very virulent plus MDV. In order to manipulate the CVRM2 genome to use it as a vaccine vector, we have generated a bacterial artificial chromosome of CVRM2. The CVRM2-BAC replicated to levels similar to CVRM-2 in vitro. The in vivo biological properties of CVRM2-BAC will be discussed.
