Interpretive Summary:

Technical Abstract: There are approximately 190 wild potato (2n = 2x = 24), tetraploids (2n = 4x = 48), and hexaploids (2n = 6x = 72), as well as a few rare triploids and pentaploids. Previous studies using nuclear ortholog DNA sequences GBSSI (waxy) and nitrate reductase (NIA) indicated that the polyploids may have autopolyploid or allopolyploid origins. The present research examined these results with six conserved orthologous set (COSII) nuclear loci in 54 accessions of 11 wild polyploid species and 23 accessions of 21 diploid species. Our results not only support part results from GBSSI and NIA, but also show some new ones: 1) some accessions from the same species have originated from different parents, 2) some accessions have gained or lost their alleles while other accessions from the same species still kept them, 3) some alleles from the polyploids have clustered together away from the diploid accessions, making the cladistic structure more complex and the origin of some polyploids unclear. These results indicate that different genomic loci give different evolutionary histories.
