Interpretive Summary: Determining the sperm numbers in a semen sample gives a measure of semen quality and is necessary if the semen is to be inseminated based on the numbers of sperm per insemination. The hemocytometer allows the investigator to could the number of cells directly from a grid. To use instruments determining sperm concentration, one must calibrate a given reading for that instrument to actual sperm numbers. This task is described in another chapter. The third chapter describes the use of one such instrument and how to derive actual sperm numbers. The chapters are geared toward both poultry industry and poultry research personnel.
