Technical Abstract: The internationally recognized Agricultural Research Service (ARS) Culture Collection will be described to include the microorganisms maintained by the collection, preservation methods and worldwide distribution of cultures. The impact of the germplasm will be described to include discovery of the first production strain for large-scale biosynthesis of penicillin, as well as strains for production of riboflavin, xanthan gum, dextran and discovery of the first yeast capable of fermenting D-xylose, a major component of biomass used in production of biofuels. The use of molecular methods for strain characterization will be discussed to include food spoilage and medically important yeasts, bacterial food borne pathogens and plant pathogens.
