Interpretive Summary:

Technical Abstract: The production of hybrid-onion seed is dependent on cytoplasmic-genic male sterility (CMS) systems. For the most commonly used CMS, male-sterile (S) cytoplasm interacts with a dominant allele at one nuclear male-fertility restoration locus (Ms) to condition male fertility. We are using proteomics to reveal differences for mitochondrial proteins in male-sterile (S msms) versus restored male-fertile (S MSms) plants. We isolated mitochondria from each population, extracted proteins, and ran 2D gels. Differentially stained spots were partially sequenced using tandem mass spectrometry and matches to mitochondrial proteins identified. The alpha subunit of the ATPase complex showed clear differences and had greater expression in male-fertility-restored umbels than in male-sterile umbels. This observation is consistent with the previously published Northern analyses of Sato (TAG 96:367), in which the transcript of ATPase a shows higher expression in male-fertility restored versus male-sterile onion. We are verifying these expression level differences using quantitative RT-PCR and Western blots.
