Interpretive Summary: This book chapter is for the sunflower volume in the Crop GGB (Genetics, Genomics and Breeding) Book Series. The book includes chapters covering basic information about the sunflower plant, germplasm diversity, classical genetics and traditional breeding, genome mapping, regulation of seed oil content, DNA marker-assisted breeding, gene cloning, and the future of sunflower crop. This chapter starts with a brief history of mapping efforts of using morphological and biochemical markers, then describes all DNA-based Marker Techniques used in sunflower genome mapping including RFLP (restriction fragment length polymorphism), RAPD (random amplification of polymorphic DNA), AFLP (amplified fragment length polymorphism), DALP (direct amplification of length polymorphism), SSR (simple sequence repeat), TRAP (target region amplification polymorphism) and SNP (single nucleotide polymorphisms). This chapter also reviews the mapping populations developed and the mapping software packages available and summarizes the sunflower genome mapping achievement of approximately 20 published linkage maps. The chapter ends with listing the challenges to the sunflower research community in three areas: 1, handling the enormous amount of data of high-throughput sequence-based markers in sunflower genome mapping in the near future, 2. correlating DNA sequence alterations with the quantitative variation of economically important traits; and 3, applying the mapping achievement to sunflower improvement, the ultimate goal of genome mapping.
