Interpretive Summary: Technical Abstract: Cathepsin H and L, a lysosomal cysteine endopeptidase of the papain family, are ubiquitously expressed and involve in antigen processing. In this communication, the channel catfish cathepsin H and L transcripts were sequenced and analyzed. Total RNA from tissues was extracted and cDNA libraries were constructed by the rapid amplification cDNA end (RACE) method. The gene-specific primers in conjunction with the RACE primers were used to PCR amplify 5’- and 3’-ends of the cathepsin H and L transcripts. The complete channel catfish cathepsin H cDNA comprised 1415 bp including a 978-bp open reading frame, which potentially encodes a putative protein of 326 amino acid residues with calculated molecular mass of 36.6 kDa and pI of 5.51. The complete channel catfish cathepsin L cDNA comprised 1639 nucleotides including a 1008-bp open reading frame, which potentially encodes a putative protein of 336 amino acid residues with calculated molecular mass of 38.1 kDa and pI of 5.84. |
