Technical Abstract: Assessing the total dietary intake of vitamins from all sources, including dietary supplements and fortified foods, would be aided considerably by having rapid and accurate analytical methodologies which are capable of simultaneous determination of several vitamins. The individual vitamins have unique solubility characteristics and susceptibility to degradation that make such an approach challenging. Vitamins naturally present in foods may occur in different chemical forms, the levels in a given food item may range over several orders of magnitude, and food matrices may also present a variety of complicating factors. Vitamins in dietary supplements and fortified foods, however, are typically added in a single chemical form, and matrix issues are usually not as complex. These sources should thus be relatively amenable to approaches that aim for simultaneous determination of multiple vitamins. Recent work in our laboratory has focused on development of LC-UV/fluorescence and LC-MS methods for the simultaneous determination in supplement tablets and in fortified foods of several water-soluble vitamins including: thiamine, niacin, pyridoxine, pantothenic acid, folic acid, biotin, riboflavin, and ascorbic acid. LC-UV/fluorescence determinations are made using external standards. LC-MS determinations are in most cases made using stable isotope dilution mass spectrometry (IDMS).
