Interpretive Summary: A mass spectrometry method was developed to identify and quantify the regiospecific isomers of triacylglycerols in plant oils and animal fats. Regiospecific isomers are different on the locations of the fatty acids on the glycerol backbone of triacylglycerols. The identification of regiospecific isomers can help to determine the biosynthetic pathway of triacylglycerols. The regiospecific isomers affect the physical properties of the oil (triacylglycerols) for industrial uses, e.g., viscosity, pour point, melting point and crystal structures. It also affects the human absorption of oil in food and is thus valuable in nutritional value for food industry.

Technical Abstract: The MS3 method was developed to estimate the contents of regioisomers of TAG, AAB and ABA, from biological origins. The content of AAB in (AAB + ABA) is estimated as (A + rA)/(A + B). A is the abundance of the ion [AAB + Li - ACOOH - A'CH=CHCOOH]+ from AAB alone. Minor amount of AAB can be detected by this method because ABA could not contribute to this ion. B is the abundances of the ions [ABA + Li - ACOOH - B'CH=CHCOOH]+ and [AAB + Li - ACOOH - B'CH=CHCOOH]+ combined at the same m/z value. r is the ratio of the abundances of the ions [AAB + Li - ACOOH - B'CH=CHCOOH]+/[AAB + Li - ACOOH - A'CH=CHCOOH]+ from AAB. The method can target the specific m/z of ABA and AAB in a biological sample and can thus avoid the contamination of ions from other TAG molecular species.
