Technical Abstract: We are completing a project to increase the diversity of EST sequences for Fragaria. Of the 19,000 Fragaria EST sequences currently in Genbank, over 9,000, from a cold-stressed seedling library, were submitted by our project. An additional 32,000 sequences will be submitted imminently. Our project focused on identifying genes expressed in Fragaria abiotically stressed tissues in order to increase the number of sequence from stress-related genes in the family Rosaceae. We have sequenced approximately 10,000 cDNA clones from each of 5 libraries from multiple tissues of plants treated with cold, heat, salt, drought, and a combination of salt and heat, and a total of 41,000 high quality sequences have been generated. All libraries have been constructed in Gateway vectors allowing multiple downstream applications. We used a diploid species of strawberry, Fragaria vesca, a useful model system for the Rosaceae. This plant has a small genome size (~200 Mbp), and a generation time of about 4 months from seed to seed. It is efficiently transformed and inbred lines are available. Our selected genotype (PI551574; Hawaii 4) is day-neutral. F. vesca transcript assemblies (TA) are now available at http://plantta.tigr.org. ESTs from F. vesca as well as F. x ananassa were separately clustered and assembled. The F. vesca TA was built from ~14,000 ESTs, resulting in ~2,000 contigs and ~6,000 singletons. The ~5,000 F. x ananassa ESTs resulted in a TA consisting mostly of singletons. Analysis of these sequences and the complete set of assemblies will be presented.
