Technical Abstract: A microbial strain has been developed that can produce xylitol from L-arabinose at a high yield by transforming Escherichia coli with a new xylitol biosynthetic pathway consisting of L-arabinose isomerase, D-tagatose 3-epimerase, and L-xylulose reductase. An E. coli strain that heterologously expressed these enzymes produced 3.1 g/L xylitol with the yield of 0.66 g xylitol/g consumed L-arabinose. Moreover, utilization of glycerol as a co-substrate significantly improved the xylitol production and yield. The E. coli strain produced 10 g/L xylitol using 10 g/L L-arabinose and 15 g/L glycerol with the yield of 1.1 g xylitol/g consumed L-arabinose.
