Interpretive Summary:

Technical Abstract: Previous reports have shown that furanocoumarins and coumarins from grapefruit juice (GFJ) are primarily responsible for the inhibition of intestinal cytochrome P450 3A4 (CYP3A4). This enzyme is responsible for the metabolism of some widely prescribed drugs, and its inhibition by these GFJ compounds can cause unexpected physiological responses. Due to the potential pharmacological actions of such (furano)coumarins in humans, an effort is underway to characterize their structure/function relationships, relative to protein binding and enzyme inhibition. Grapefruit oil wax (GFW) is a sub product of grapefruit peel oil (GFO) and contains a complex concentrated mixture of compounds, some of them found in GFJ. In this study we isolated new components – furanocoumarins and coumarins – from GFW and described their spectral characteristics. These compounds were also tested for their inhibition of CYP3A4 relative to known standards such as bergamottin and 6',7' dihydroxybergamottin.
