Interpretive Summary:

Technical Abstract: We screened by PCR a doubled haploid broccoli Early Big BAC library for genes CYP79F1, CS-Lyase, S-GT, and GS-OH using BAC pools. For that purpose, we used primers based on conserved regions of Arabidopsis thaliana orthologs. We designed the primers from the Brassica database to amplify the whole sequence of these four genes in the identified BAC clones of broccoli. The four BACs harboring these genes have been sequenced and the four genes have been mapped in two mapping populations. Previously, we mapped three other major genes, BoGSL-ALK, BoGSL-ELONG and BoGSL-PRO. Therefore, altogether we have now mapped seven major genes in this pathway. The QTL analysis has been performed in these two populations. Generally, these genes except S-GT and CYP79F1 had been involved in the QTL region with the corresponding traits. Additional QTLs had been identified for corresponding traits showing the complexity of the pathway. Many significant interactions have been detected which reveal the close relationship of different QTLs in the genome.
