Interpretive Summary:

Technical Abstract: The objective of this research is to use microscopy methods to determine whether chemical methods used to describe the degree of milling (DOM) represent “surface” or “total” lipids. This presentation serves to update the progress of our research using microscopy to our collaborators at the University of Arkansas, other ARS agencies doing rice research and the industry partners. Lipid bodies were located on and in the rice kernel using fluorescence microscopy both before and after various degrees of milling as well as after the surface extraction of lipid. Lipids were located using nile blue A, the active component of which is Nile Red, which fluoresces most intensely with neutral lipids. Other microscopy methods will be used to describe and correlate surface (scanning electron microscopy) and interior (light microscopy) changes resulting from milling and extraction can be described.
