Technical Abstract: A complete catalogue of the human complement of salivary secretory components is critical to understanding oral health and nutrition. Recently there has been an interest in exploring the whole glycome of human saliva using hyphenated chromatography-mass spectrometry techniques. In addition to over 2000 proteins, saliva contains a diverse array of low molecular weight peptides, lipids, carbohydrates, steroids, glycolipids, and electrolytes. Characterization of these small molecules poses a challenge due to their low abundance. Here we employ LC-MS/MS and GC-MS to determine the identity of the small molecules. The low molecular weight (< 3 kD) fraction of parotid saliva was collected from a healthy saliva donor and Bligh-Dyer fractionated. The hydrophobic components were subjected to LC-MS/MS analysis using a standard reverse phase column and the hydrophilic components were subjected to LC-MS/MS analysis using an amino column. The carbohydrate components (as aldononitrile acetates) and lipids (as FAMES and pyrrolidides) were further analyzed by GC-MS. The elution time, mass value, MS/MS and EI fragmentation patterns provided a catalogue of these small molecules.
