Interpretive Summary:

Technical Abstract: Although the commercial aquaculture of hybrid striped bass ranks fourth among finfish in the US in economic impact, the industry still relies heavily on wild-caught stocks for fingerling production. The construction of genetic markers for Morone species will facilitate the development of domesticated broodstock through selective breeding. To this end, we developed 345 microsatellite markers for population genetic studies and genome mapping. Microsatellite repeat-enriched libraries (AAG, ATG, TACA, TAGA) were used to identify candidate sequences for marker development. A subset of 72 markers was selected for characterization by genotyping across a panel of fish from diverse origins. Fifty-two markers were amplified successfully and found to be polymorphic, with 2-18 alleles and PIC values ranging from .2 to .87. Five markers were observed to amplify multiple loci as would be observed as a result of a genome duplication event. One of these markers had sufficient distance between allele sizes such that it was analyzed as two markers.
