Interpretive Summary: An important gene related to energy metabolism and fat deposition was studied. The gene, Adiponectin (ADIPOQ), is a plasma protein that regulates energy usage via glucose (blood sugar) and lipid (fat) metabolism. A portion of ADIPOQ was sequenced and the gene was placed on the pig genome map using two independent methods. Both methods assigned the gene to pig chromosome 13 which is consistent with where it maps in the human genome and the current comparative pig-human map. This gene is a candidate gene for fat deposition and its assignment improves the pig-human comparative map.
Technical Abstract: Adiponectin, an adipose tissue-specific plasma protein, is an adipocytokine that regulates energy homeostasis, glucose and lipid metabolism. A portion of porcine ADIPOQ (adipocyte, C1Q and collagen domain containing) gene encoding for adiponectin was amplified and sequenced. Sequence of 3326 bp encompassing intron 1, exon 3 and intervening intron 2 and exon 2 has been deposited in the EMBL/GenBank/DDJ databases (AJ849536). The mRNA sequence deduced from AJ849536 matched the human mRNA sequence NM_004797 and already deposited porcine mRNA sequences NM_214370 and AY589691 with the identities 85% (584 of 680 nucleotides), 98% (668 of 677 nucleotides) and 100% (768 of 768 nucleotides), respectively. A length polymorphism was genotyped in the USDA-MARC resource population to determine ADIPOQ’s location on the genetic map. Multipoint linkage analysis based on 53 informative meioses placed the ADIPOQ gene on the current USDA MARC chromosome 13 linkage map at position 53.6 cM. Radiation hybrid mapping in the INRA-University of Minnesota porcine Radiation Hybrid (IMpRH ) panel concurred with linkage results. Human gene was mapped to chromosome segment 3q27. Our results are in agreement with comparative mapping data.
