Technical Abstract: Technical Abstract: Citrus leprosis disease has been present in several South American countries for many decades. The recent appearance of leprosis in Central American countries poses a serious threat to the U.S. citrus industry, where it does not occur. Cytoplasmic- and nucleo-Rhabdovirus-like particles have been associated with leprosis. Screening and sequencing of clones from a cDNA library made from cytoplasmic-leprosis infected tissue has lead to the identification of putative viral sequences. RT-PCR analysis and Northern hybridizations have shown consistent association of this sequence with leprosis infected trees having cytoplasmic virus particles from several countries, but not with samples from leprosis infected trees having the nucleo-virus particles or with healthy citrus. Northern hybridizations by using DIG-labeled DNA and RNA (sense and antisense) probes of different clones have shown two different patterns. Probes from clones from contig 1 hybridized with two RNAs of approximately 10 kb and 1.2 kb, while probes from clones from contig 2 hybridized with four RNAs of 4.7, 2.5, 1.6, and 1.0 kb, respectively. These results clearly suggest that at least two species of RNAs are associated with the virus causing leprosis disease and that the cytoplasmic- and nucleo-type of leprosis are caused by two distinct viruses.
