Interpretive Summary:

Technical Abstract: The cytokines regulating the T helper type two (Th-2) responses in swine have not been clearly defined. Studies have indicated that IL-4 is seldom detected in the T cell response to antigen or mitogen when analyzing peripheral T cells of swine. It is known that IL-4 and IL-13 are important cytokines in the Th-2 response in mice and humans and often have redundant function. The objective of this study was to analyze the function of IL-13 in swine. The porcine IL-13 gene was cloned and expressed in a replication defective adenovirus vector, Ad5blue. Gene-specific anti-peptide sera to the amino- and carboxi- termini of the protein were used to identify the recombinant product and to detect the IL-13 protein in biological samples. We have successfully expressed the recombinant porcine IL-13 in the recombinant adenovirus vector. The protein has been shown to be biologically active in combination with porcine GM-CSF inducing differentiation of peripheral blood monocytes into immature dendritic cells. These data indicate that in the absence of IL-4 in the periphery of swine, porcine IL-13 mediates important effector functions attributed to IL-4 in other species.
